Venusaur is a really nice Chlorophyll sweeper usually but you can probably make it work. Vileplume just has the niche perks of Strength Sap and one of the only mons that runs effect spore. Rocky Helm just makes her job easier and 1 Surging Strike on Vileplume will take half of its health assuming you're running physical defensive Vileplume with Rocky Helm.

Also I meant that Urisifu takes 50% health against Vileplume on Surging Strikes. Vileplume eats it quite easily. So I found it as a nice Urisifu counter casually testing it out.
Venusaur is bulkier, but doesn't have Effect Spore to punish contact moves.
Flame Orb Luna doesn't really care about Effect Spore, it's already got a status effect on it (Burn).
That aside, I can see where you're coming from. It might not be ranked there, but it could probably have a decent niche in OU.
I kinda forgot about that yeah. But even then it can reasonably tank a lot of what Ursaluna throws pre-SD and have a decent toolkit to slow it down in strength sap, sleep powder assuming it's bulletproof/turn 1 dumb play etc. It also has 110 Sp atk which isn't much for something like Vileplume but uninvested can still hurt things like Iron Valiant and can go toe to toe with Tusk/Rapid Strike.
Not saying it'll be meta defying, but it definitely has a strong niche use. Tried it on Natdex for fun and it's pretty crazy what you can get away with against huge physical offensive threats. So I'm excited to run her to prove the world that she is slept on.

Assuming those switch ins are resists, not really. And she can reasonably punch everyone listed except Guts Ursaluna with Effect spore assuming you can get it to work. And a 2HKO is good because again, you can basically middle finger the opponent with strength sap on next turn anyways if they go for one of those moves right after a resist/netrual hit.

is right, a counter is something that can directly switch into a pokemon and still win 1v1 in the worst case scenario. That being said you're forgetting a few things.
1. Vileplume is essentially a dead pokemon versus gholdengo.
This is a huge and really big problem for it, especially when Gholdengo roams wild in the tier.
2. Vileplume will take a hit and literally die according to your calcs, and the best it can get off is maybe a sleep powder. Amoongus is literally 20 times better in this scenario.
3. You listed Ursaluna, Urshifu, Sneasler, Kingambit, and Great Tusk in your calcs.
Urshifu is banned.
Great Tusk is more an offensive utility pokemon and multiple other pokemon answer it well.
Ursaluna, Sneasler, and Kingambit are mostly settup sweepers, if they Swords Dance and you strength sap vileplume is dead. If you sleep power you're basically budget amoongus that might miss.
Vileplume is just outclassed by amoongus and therefore won't see play.
To add on to all of this, those calcs are with max bulk Vileplumes. It doesn't have any offensive presence as a result and can't threaten even as a check over a counter.
