the horse god needs specific ones to upgrade your horse stats, and there's at least 2 side quests that need specific recipes. Other than that there's no point.

Aren't hearty truffles in limited quantity per playthrough?
No, they come back every blood moon. The limiting factor is there is no location that has a lot of them all together, they are spread out and only found 1 at a time. The materials for the cheesecake can be purchased in bulk so you can make as many as you want, thus alleviating the limited inventory problem.
For healing purposes? None really, like gimmick ingredients do have a pretty good duration factor for the first one of each in a recipe if i remember the BOTW mechanics compared to most other food (but only for the first one of each), though Insects and Monster Parts generally BTFO them (But don't add healing hearts). Hearty doesn't use Duration though, so it is useless for them.
Basically in theory they are filler for shit like porgies that have strong Buff strength, in practice you can just make elixirs for buffs and slap in a dragon part (easier then ever since the dragons are much easier to farm then BotW) or save scum monster extract to make it long lasting, if you even need it to be THAT long lasting and for Healing/Stamina Restoration Hearty and Enduring remains as broken as ever.

Nah, they respawn and even if they didn't there are plenty of other Hearty options as long as you make a mental note of where you found Radishes/Bass (Like the Mirror Isle once you dry it out has a couple of bass to grab, and as long as you can find one Big Hearty Radish you can literally farm them in Hateno and get two there each blood moon or so, if not more often i only checked in after Blood Moons.)
It is kinda funny since they obviously realized Hearty was broken good since they tried to make it rarer and seem to have removed Durian, but it didn't actually fix anything.
Gloom enemies also make temporary hearts not as OP. Also Ganondorf fucks you over if you rely on temporary hearts because not only does he have Gloom effect but he also has an attack that steals your hearts.
Just cut grass and hoard some crickets.
4 crickets + pretty much any monster part = Full stamina wheel potion
Also anything that gives you yellow stamina will fully replenish your green
