It really depends what kind of 100% we are talking about. If it's 100%ing the story and all side content then I'm confident I can do it. If 100%ing means to get all items, enemy skills, max all material and stats then no fricking way.
I'd say all sidequest content, but that's a tough 24 hours unless you've been practicing speedrunning for months. I would say a "complete" playthrough would have you getting all the MASTER materia and killing all the WEAPONS on top of getting everyone's ultimate weapon. Personally, I'd throw in watching all the optional cutscenes and shit, but there's no way to check your file to see if you saw some of them if you missed them so I'd skip if I were doing the challenge. I'd say the kicker is filling the Enemy Skill Materia.
These are the speedrun rules for a 100% run:
All Bosses:
Every Boss needs to be defeated(listed in the sheet)
Recruit Vincent and Yuffie:
Vincent and Yuffie need to be unlocked as playable characters.
All Limits:
All Characters(including Aerith) need to have their Level 4 Limit learned.
Sidequests:
Gold Chocobo: Breed a Golden Chocobo at the Chocobo Ranch.
Kalm Traveler: Give the Guide Book, Earth Harp and Desert Rose to the Traveler.
Turtle Paradise: Obtain the items from the Turtle Paradise barkeeper.
Wutai: Defeat Rapps and reobtain your stolen Materia
Pagoda: Defeat Godo in the Pagoda
Lucrecia: Talk to Lucrecia behind the Waterfall
Battle Square: Win the Special Battle.
Villa de Cloud: Purchase the Villa in Costa del Sol for 300.000 Gil
Huge Materia: Obtain all 4 Huge Materia from Fort Condor, Corel, Submarine and the Rocket. They don’t need to be placed into Bugenhagens observatory.
All Items(listed in the Reference Sheet):
Collect all usable Items, Weapons, Armor, Accessories.
Exceptions: Key Items, Limit Manuals, Desert Rose, Earth Harp.
All Items must be in the Inventory when finishing the game.(Exception: Save Crystal)
Materia(listed in the Reference Sheet):
Collect all Magic, Command, Summon, Support and Independent Materia.
Materia does not have to be mastered.
All 4 Enemy Skill Materia must be obtained and mastered.
All Materia must be in the inventory when finishing the game.
Speedrun Rules:

Verification Process:
When all requirements have been met the following things have to be done:
- Remove All Materia, Arrange and scroll through them
- Sort Items by “Type” and scroll through them
- Show every characters Equipment and Limits
Save Crystal can be set in the last room before the Final Descend and a save made.
After the run the save can be loaded and the run be verified as shown above.
Not saying this is impossible, but I'm certain I couldn't do it. At least not on the first try.

Yeah, I can do it.
I 100%'d that game, backtracking and all and got all achievements in 30 hours. I could easily rush to the end of the game in 10 hours or less. Probably much less. If I used glitches and abused bugs, I could probably do it in like 2 or 3.
I found a tonne of bugs in that game without looking shit up. Like infinite jump.

I have to say, it's quite manageable, I know where to find all of the powerups, so 100% shouldn't be an issue, what might give me trouble is playing on the most difficult setting, especially with bosses like Hammerhead, which are difficulty, but I can do it.
