No, those dots are absolutely safe, the combination of numbers guarantee that the mines have to be in a certain area, thus in those marked squares there's 0 chance to have a mine

I always noticed that in expert the mine in these situations is always horizontal or vertical but in intermediate it was always diagonal. The game places the mines as you play, not on gen. That's why the first click is never a mine.
