I think Sora's multiple tiers stronger than Mumei and Aki. I find it difficult to lose as Sora. I've died as Mumei and Aki before.
With Mumei she does insane DPS and cleave. Pick ranged weapon items, never any melee.
For Aki you have to focus on debuff item stamps and haste+. These are both big damage cannon characters, Mumei is single-target/cleave and Aki is AoE
I know what the characters do. I've still lost as them before.
I personally only care about how strong something is when the game is played normally.

I'm literally the only person I've ever seen post a tier list, and whenever I do, people tear it apart. They never post theirs, though.
I think most people who play this game enough to even consider a tier list are probably the obsessively le hardcore gamer types who max out upgrades and spend 99% of their playtime trying to break records, so all they really know about the game is what builds are broken.
In all honesty, tier lists hardly make sense outside of competitive and are outright redundant when every character can clear every stage with relative ease. The only tierlists that make sense for a game like this are hyper specific lists like Endless survival, Time Stage, money farming, etc.
Good characters: Takanashi Kiara, Nanashi Mumei, Sakura Miko, Ceres Fauna, Irys, Yuzuki Choco, Aki Rosenthal
Maybe you can look at the leaderboards and compare scores per character.
I looked at it back around when I picked it up and I think Choco generally had the highest scores.
I just tried her for the first time and if the charm works on YAGOOs I wouldn't be surprised. Making them kill each other seems pretty strong.
All S-tier characters have a gimmick that synergizes really really well with certain items and weapons and stamps.
Either it's huge AoE damage, huge healing ability, or huge single-target cleave

the game only has like 20 fricking items anyway who cares if a char is weak or strong it's going to feel the same as every character in every run in 10 minutes. how do people play this game for more than 5 hours.
Isaac has ruined any game with funni random item mutations because they are always weak as frick in comparison.
Game has a lot more than that since each character has 3 skills and it's not even fully released. they want to add alternate special skills for each character as well.
there are 16 weapons and 34 passive items not counting fusions. Additionally there builds like purposely sealing weapon drops to focus on passive stat ups.
Yuzuki Choco is the most busted in the game if you get +food item drop armour, the +life drain stamp, the sake item and basically just stack as much healing items / weapons / stamps / etc. as you possibly can along with crit+, haste+ and the most important thing of all, Full Meal (doubles healing), because every time she heals she can crit and those heals when they crit do 400% (technically 800% with the Full Meal item) as damage to enemies within 200px of her. It's insane how much healing and DPS she can do, just try with what I mentioned.
Pro tip if you want to be lazy and farm coins, the fishing game gets you mad stacks

Stack speed buffs. One of her skills applies an ATK buff from her SPD stat, so pick up Energy Drink and SPD buffs whenever you can. Also, close range attacks (Spider Cooking, BL Book) and collab those for close range attacks.
She's the only one who can outrun the Yagoo swarm at the end of the stage, I've found. Not sure how long she can keep at it, though.

How do I build Irys?
Her kit has defensive skills already just focus on attack items.
Crit.
Her whole build revolves around getting crits, which gives her heals, which gives her bonus to attack. Sake, Hope Soda, and GWS Pill are the best choices. Her main weapon is also excellent, and will chew through nearly everything once at full power. Depending on how risky you want to play, you can also pick up Face Mask, Energy Drink, and even Injection Asacoco (haven't tried that one) or you can grab Headphones (to complement her skill which knocks enemies back), Body Pillow, and Chicken Feather for more defenses.
Super Chatto Time is amazing if you get the stamp which drops coins when you attack things, since you vacuum them right up.
As for other weapons, she doesn't really need many. But the Dragon Fire collab helps her cut through enemies even more and Broken Dreams/Frozen Sea collab slowing everything down so they're easier to chew through.
I know nothing of Vtubers but I think Aki is my favorite because she can instant kill bosses via belly dance.
I keep mixing up Aki and AZKi's skills because they're both so reliant on running around for big dick damage.
