Those characters have a single beauty mark in the typical Marilyn Monroe fashion, they aren't designed with blotchy freckles and moles all over their skin. That's something western fan artists stereotypically add in their own interpretations of the characters, like making characters fat, brown and/or having a bright red nose for some reason. 2b is a good example- she doesn't have freckles, she's a robot and wouldn't get freckles from sun exposure, but the westerner feels the need to impulsively coat her in freckles anyway.

I’m pretty sure the appeal is in the implication that the character isn’t wearing makeup. Nothing is left to the imagination on how how her face is flawed.
A lot of popular female characters are also drawn with scars that usually infer something to the audience the same way hair color is linked to a lot of personality tropes.

that's a very western perspective, Japanese fans are well aware of the western freckle trend and view them the same way you do the rest of the things you mentioned- unnecessary blemishes. Their ideal is pale, clean skin. A single mole to denote a sultry mature woman is generally the most you'll see on a character that's meant to be attractive.
